Description

Hexarelin is a synthetic peptide composed of six amino acids, structurally related to classical GHRPs but with modifications conferring greater metabolic stability and higher affinity for the GHS-R1a receptor. In acetate form, the peptide exhibits enhanced physicochemical stability for storage and laboratory handling.

Its mechanism of action is based on direct activation of the growth hormone secretagogue receptor, expressed in the hypothalamus and anterior pituitary. Receptor activation triggers intracellular signaling cascades associated with increased calcium influx in somatotroph cells, culminating in pulsatile GH release.

Unlike GHRH analogs, Hexarelin acts through a pathway independent of the GHRH receptor, enabling complementary GH secretion stimulation. The peptide exhibits a short half-life, though superior to other classical GHRPs due to greater resistance to plasma peptidase degradation.

Hexarelin’s biological activity depends on the functional integrity of the hypothalamus-pituitary axis, neither acting as exogenous hormone nor replacing endogenous GH secretion.

What is it

Examorelin (INN) (development code names EP-23905, MF-6003), also known as Hexarelin, is a potent, synthetic, orally active, centrally-penetrant, and highly selective agonist of the ghrelin/growth hormone secretagogue receptor (GHSR) and a growth hormone secretagogue developed by Mediolanum Farmaceutici.

It is a hexapeptide with the amino acid sequence His-D-2-methyl-Trp-Ala-Trp-D-Phe-Lys-NH2 derived from GHRP-6 (which in turn is a ghrelin analog).

Examorelin dose-dependently increases plasma growth hormone (GH) levels in animals and humans.

Additionally, similar to pralmorelin (GHRP-2) and GHRP-6, it slightly and dose-dependently stimulates the release of prolactin, adrenocorticotropic hormone (ACTH), and cortisol in humans.

There are contradictory reports on examorelin's ability to elevate insulin-like growth factor 1 (IGF-1) and insulin-like growth factor binding protein type 1 (IGFBP-1) levels in humans; some studies found no increase while others found a slight but statistically significant increase.

Examorelin does not affect plasma glucose levels, luteinizing hormone (LH), follicle-stimulating hormone (FSH), or thyroid-stimulating hormone (TSH) in humans.

Examorelin releases more GH than growth hormone-releasing hormone (GHRH) in humans and produces synergistic effects on GH release in combination with GHRH, resulting in "massive" increases in plasma GH levels even with only low doses of examorelin.

Doping: Prohibited substance - Ordinance No. 411/2015, of November 26 - Approves the List of Prohibited Substances and Methods for 2016 and revokes Ordinance No. 270/2014, of December 22.

Common Uses
Hexarelin stimulates growth hormone release by activating the ghrelin receptor found in the brain.
Activating this receptor and increasing growth hormone levels in the body can modulate food intake and energy metabolism, as well as influence glucose and fat metabolism.
Growth hormone secretion also increases appetite and causes muscle growth.
